Dier misses out with a muscle injury, with Hudson-Odoi set to be trialled in Sancho's place for the Euro 2020 qualifier.
Meanwhile, Jordan Henderson is set to earn his 50th Three Lions cap alongside Ross Barkley and Rice in midfield.
Jordan Pickford is set to retain his spot between the sticks, with Southgate making changes in the full-back slots.
Kieran Trippier and Danny Rose could come in for Kyle Walker and Ben Chilwell in a fresher-looking XI.
DISCIPLINE IS KEY
However, Manchester City star Walker is the only player in the current squad to have played in Montenegro.
In an atmosphere expected to be hostile, Southgate could yet stick with him at right-back for some much-needed poise and experience.
Ahead of the game, the England boss demanded discipline from his team in what will be a tougher test than the Wembley stroll.
Southgate said: "I know the passion the supporters have for their country and of course that gives a huge lift to their own team.
